EXTRACTION OF GENOMIC DNA FROM JATROPHA sp. USING MODIFIED CTAB METHOD
نویسندگان
چکیده
Genomic DNA was isolated from leaves of three species of Jatropha, namely J. glandulifera, J. gossypifolia and J. curcas. The objective of our study was to use alcohol as a fixing solution, making liquid nitrogen unnecessary for isolation of genomic DNA from Jatropha species. The spectral quality of genomic DNA isolated using this method as measured by the A260/A280 absorbance ratio ranged from 1.83 to 1.94 for all three species of Jatropha. DNA quality and quantity were comparable to those isolated with liquid nitrogen. The purity of the isolated DNA was further confirmed by PCR (RAPD) using 4 decamer primers. DNA samples prepared by this method were consistently amplifiable in the RAPD reaction and gave reproducible profiles. This method does not require for fixation or grinding in liquid nitrogen, making it advantageous over common protocol.

Isolation of Genomic Dna from Five Species of Euphorbiaceae without Liquid Nitrogen
The isolation of intact, high-molecular mass genomic DNA is essential for many molecular biology applications including PCR, endonuclease digestion, southern blot analysis, and genomic library construction. Many protocols are available for the extraction of genomic DNA from plant tissue, and all the protocols have used expensive and hazardous chemical-liquid nitrogen for grinding plant material...
متن کاملAn efficient and simple CTAB based method for total genomic DNA isolation from low amounts of aquatic plants leaves with a high level of secondary metabolites
An efficient DNA isolation protocol specifically modified to get pure quality DNA required for molecular studieshas been reported in this paper. Some aquatic plants (Potamogeton spp., Ceratophyllum demersum and Myriophyllum spicatum) were used for the study. The protocol developed will be useful in getting high and pure DNA. Instead of using the available DNA extraction kits, this protocol can ...
متن کاملOptimization of the genomic DNA extraction in some mosses
The presence of organic compounds and high amount of secondary metabolites (polysaccharides, phenolic component, etc.) in mosses cause difficulties in DNA extraction that are followed by problems in PCR reactions. In lower plants, various methods have been used for DNA extraction including silica gel and different commercial kits. These methods mostly use hazardous (like phenol or liquid nitrog...
متن کاملA simple and efficient DNA extraction protocol for old herbarium leaves of Bellevalia (Asparagaceae, Scilloideae)
High-quality DNA extraction plays an important role to make sharp bands in the gel electrophoresis and also produces clean chromatograms. Usually, DNA extract is delivered using the modified CTAB method but this method cannot obtain high-quality DNA for molecular analysis from old dried leaves of Bellevalia due to having different chemical compounds which inhibit to obtain a clear DNA extractio...
متن کاملA simple, rapid and efficient method for the extraction of genomic DNA from lychee (Litchi chinensis Sonn.)
A simple, rapid and efficient method for isolating genomic DNA from lychee (Litchi chinensis Sonn.) was developed. This modified CTAB protocol include the use of 2 M NaCl, PVP, 5% mercaptoethanol and 80% ethanol in the extraction as well as reducing the centrifugation times during the separation and precipitation of the DNA. Isolated genomic DNA showed high purity and high quantity following re...
